Cell Proliferation Assay Services

Cell proliferation assays are a cornerstone of stem cell and cancer cell pathway analysis and drug testing for efficacy and safety.  Medicilon is a cell proliferation assay services platform that provides a rapid, sensitive and economical way to quantitatively measure cell proliferation and cytotoxicity in various human and animal cell lines.

Cell proliferation, also known as cell growth, cell division, or cell replication is the basic process through which cells form new cell. Cell proliferation is the increase in cell number as a result of cell division and growth. The process of cell proliferation depends on the prior occurrence of a series of internal cellular changes before either mitotic or meiotic division starts. These changes occur in a given orderly sequence known as the four phase of the cell cycle, and both mitosis and meiosis are the final steps of a cell cycle.

The development of assays for measuring the survival of individual tumor cells revolutionized the study of experimental cancer therapy by enabling researchers to move from assessing the gross responses of tumors to measuring the survival of cells in the critical, clonogenic tumor cell population. The development and the use of these assays formed the basis for many of our modern concepts of tumor biology, from the concept of logarithmic cell kill to considerations of cell proliferation kinetics.

The accurate assessment of cell number and cell proliferation is useful in many high content assays and is a key read out in cytotoxicity and apoptosis applications. Proliferation assays such as MTT, XTT and WST-1 were commonly used for measuring biocompatibility. Enzyme-based methods using MTT and WST -1 is a reductive coloring agent and dehydrogenase in a viable cell to determine the cell viability with a colorimetric method. This method is far superior to the previously mentioned methods because it is easy – to –use, safe, has a high reproducibility, and is widely used in both cell viability and cytotoxicity tests. Therefore, this method is suitable for those who are just beginning such experiments. Among the enzyme based assays, the MTT assay is the best known methods for determining mitochondrial dehydrogenase activities in the living cells.

These methods are limited by the impracticality of processing large numbers of samples or by the requirement for handling hazardous materials. The Calcein-AM Cell Viability Kit, XTT and MTT Cell Proliferation Assays provide rapid, versatile and accurate methods for quantifying cell viability in standard culture conditions. 3D Culture Cell Proliferation Assays are available that provide a more physiologically relevant assessment of stem cell proliferation within a 3D extracellular matrix.

MTT Cell Proliferation Assay
MTT Cell Proliferation Assay is a sensitive kit for the measurement of cell proliferation based upon the reduction of the MTT tetrazolium salt. In actively proliferating cells, an increase in MTT conversion is spectrophotometrically quantified. Conversely, in cells that are undergoing apoptosis, MTT reduction decreases, reflecting the loss of cell viability.

Calcein-AM Cell Viability Kit
The Calcein-AM Kit provides a simple, rapid and accurate method to measure cell viability and/or cytotoxicity.  Calcein-AM is a non-fluorescent, hydrophobic compound that easily permeates intact, live cells. The hydrolysis of Calcein-AM by intracellular esterases produces calcein, a hydrophilic, strongly fluorescent compound that is well-retained in the cell cytoplasm. Cells grown in black-walled plates can be stained and quantified in less than two hours.

XTT Cell Proliferation Assay
The use of XTT tetrazolium salt to assay cell proliferation, cell viability and/or cytotoxicity is a widespread, established practice. The procedures avoid radioactivity, allow for rapid determination in microplates and give reproducible and sensitive results. XTT, a yellow tetrazolium salt, is cleaved to a soluble orange formazan dye via the succinate tetrazolium reductase system in the mitochondria of metabolically active cells, which can be measured by absorbance in a microplate reader. This kit includes both XTT and the electron coupling reagent for a convenient and simple assay.
